If I put a ripped movie into iTunes, will iTunes make it's own copy of the movie file? ie, can I delete the ripped version from my hard drive and be left with only the copy in iTunes?

If you have "Copy files to iTunes Music folder when adding to library" checked, then it will have its own copy. You can then delete the copy that is outside of the music folder.

I would do a quick Get Info on the particular movie (in iTunes) and double-check the "Where" under Summary to make sure you are deleting the right one.
