Since I have two iTunes programs sharing a single media file on an external hard drive I have had to move the iTunes Music folder on both machines. If iTunes is pointed at the folder is there anyreason to actually keep and itunes music folder in my music on either machine?

There shouldn't be any reason to keep two copies, you can delete one AFTER you've confirmed that all your files are in the other folder and that the two machines are both using that particular folder. iTunes is made to be able to share a music library among multiple users (in addition to streaming music, which is also called "Sharing").
