Is there a way to automatically block a prank caller's number with the iPhone 4?

Nope. You can add the caller to a "Spammer" contact and then set the ringtone to a silent ring.

Some cellular carriers also provide call management features that will allow you to block certain callers, however they usually charge an additional fee for this.
You can also use the "Do Not Disturb" feature in iOS 6 to only allow certain callers, blocking anybody who isn't in your contacts or favourites, but this is kind of the reverse of what you're looking to do and will have the disadvantage of also disabling all other notifications.
