VirtualDubMod + Xvid + yamb
Install xvid, setup VirtualDubMod to deinterlace with blend setting and then resize (to 320 x 240) with lanczos3, set xvid to 2500kbs in SP mode, encode the video, use yamb to extract the video and then lpcm audio from the avi, then put the audio though itunes and convert to 160kbs aac (this will not work with ac3 audio) then go back to yamb and then mux the video and aac audio in to a .mp4 file.
Done.

Nothing is Best, since it depends on the user and its requirements.
But HandBrake is really good. :)

Handbrake IS the best for Mac users.
