I have a single data point from my own experience: I found them very uncomfortable, to the point of hurting my ear canal. Dull pain lingers for several days. They are sitting un-used at the moment, waiting for a better fit solution (tri-flange?) :(
I am entirely certain that comfort vaires from person to person, depending upon the size and structure of your ear canal. I have seen many others report no issue with comfort whatsoever.
As for sound quality, these things are simply amazing! ...I just wish I could wear them.
Good luck!
There is a review of the Shure E2s in the review section on this forum. It is a good read.
I have a pair and they do take some getting used to. Not the easiest to get in there but do sound better than the Apple phones. I have to ride a bus on campus and the noise was very loud, I would have to have the ipod cranked. I can now listen at a lower volume with the Shure phones.
Go to their (Shure's) website and read up as there are different versions that offer different fits if you want to spend more money for a custom mold. I wound up using the foam orange ones intsead of the clear dome ones as they fit better.
I would recommend them but I keep realizing one thing. They are best if you listen for extended periods of time and do not have to keep putting them in and taking them out - that gets old.

They are comfortable to wear....you just have to experiment with them to get the best fit for you.
For me, the plasic earpieces were a BIT** to wear. The pain after 2-3 minutes were almost incapacitating. I once had to stop walking, SLOWLY and CAREFULLY pull them from my ears because they hurt so much. I switched to the foam earpieces and found heaven with these headphones. Now it feels like i'm sticking a plush Q-tip into my ears. They sound great and they are comfortable enoegh to exercise with.
The only drawback i have is the sound isolation. It isn't really a drawback if you have had experience with sound isolation or if that is what you want. I just wasn't use to it the first month. Putting these on and walking around campus was like blocking out the rest of the world. I am very cautious about wearing the E2s around friends because i could NEVER hear them talking to me. They always have to tap my shoulder or wave at me to get my attention. It makes one feel like your being antisocial. :) I've grown to love the isolation, especially for studying. However, i wouldn't recommend using these while road biking, joggind on busy streets, or any other activity wear your hearing is vital to your safety.

I find the mine with the foams very comfortable. They just disappear.
But they are not good if you need to take them in and out a lot. It looks a little weird putting them in in public and it will bother the ear canal.

I use the small plastics that come with shure E2, it takes time getting used to these earbuds, sound quality is excellent.
